This composite part is our kill switch. P-gsiB is a salt inducible promoter and P-atp2 is an alkali inducible promoter. LacIq is an enhanced repressor in lac operon that can reduce the basal expression. Tac promoter(Ptac) is an inducible promoter that can bind to LacIq and therefore be inhibited. NdoA is an endoribonuclease toxin that is toxic for cell growth.
At high saline or alkaline or both, the lacIq behind Pgsib and Patp2 will express and the repressor will bound to tac promoter to inhibit the expression of the toxin ndoA. At low saline and alkaline, there is no longer repression and ndoA will express, leading to the death of bacteria.
We designed this kill switch to make sure that our engineered bacteria suicide when salt and pH level of the saline-alkaline soil fall back to normal.
